Anti-EGFR Aptamer targeted biopolymer nanocomposite containing Noscapine for cancer cell therapy

Cancer, as one of the leading causes of death, has led to momentous adverse impacts on human beings’ life. As a result, its diagnosis and therapy are contemplated as a major issue faced with human health. In this study, novel carboxyl modified (C) and single-strand DNA-aptamer (Apt) functionalized Chitosan (CS) nanoparticles were effectively developed for targeted delivery of noscapine (NO) and Fe 3 O 4 nanoparticles to MCF-7 breast cancer cells. The main aim of the work was breast cancer cell diagnosis and therapy. After synthesizing Apt-C-CS-NO-Fe 3 O 4 nanocomposite, analytical devices such as FT-IR, DLS, and TEM were used for its qualification and quantification. The size range was determined between 15 nm to 50 nm in diameter. Thereafter, biomedical tests, including cell viability and cell uptake iron content were done for evaluating nanocomposite’s safety and delivery. Based on results, MTT test results were shown between 75% and 100% cell viability for C-CS, Apt-C-CS, and Apt-C-CS- Fe 3 O 4 samples after 24 h and 48 h of treatment with 5 µg/mL and 10 µg/mL while it was from 65% to 70% for NO-containing nanocomposite. The iron contents of cancer cells treated with Apt-C-CS-NO-Fe 3 O 4 and Apt-C-CS-Fe 3 O 4 were approximately two folds higher than that of C-CS-Fe 3 O 4 sample. In vitro cell analysis for Apt-C-CS-NO-Fe 3 O 4 nanocomposite which further approves the importance of targeted attachment and drug delivery.
