Characterization of Collagen Extracted from Chicken and Duck Eggshell Membrane

The source of collagen which is widely used comes from animal sources. It is necessary to look for sources of collagen from other materials. The objective of this research was to analyze the characteristics of collagen extracted from eggshell membranes (chicken and duck) using papain enzyme with extraction time (6, 12 and 24 hours). The research used a Completely Randomized Factorial Design and each treatment was repeated three times. The observed parameters were physicochemical characteristics of collagen (water content, ash, protein, yield, pH, solubility in acid, color (L*, a*, b*, whiteness index), FTIR, and SDS-PAGE). The results showed that eggshell membrane had significant effect on yield, ash content, protein, L*, and b*. Extraction time had significant effect on yield, ash content, protein, pH, L*, b*, and solubility in acid. The interaction of those treatments had significant effect on yield and protein. The eggshell membrane collagen of chicken and duck has a protein structure of α1, α2, and β. The main absorption bands in the collagen were amide A, B, I, II, and III. The best treatment based on the highest value of yield, protein, whiteness index, solubility in acid, and low ash content of collagen from duck eggshell membrane with 24-hour extraction time. The characteristics of the best treatment were yield 1.78%, protein 80.78%, whiteness index 60.07%, solubility in acid 91.00%, and ash content 0.85%.
