ISM2 Is a Novel Prognostic Biomarker and Correlates with Tumor Immune Microenvironment in Colorectal Cancer

Background and Objectives: Isthmin 2 (ISM2) belongs to a family of secreted proteins. Its function and roles in tumor pathogenesis remain largely unknown. We intend to investigate the potential biological roles and clinical importance such as immune therapy of ISM2 in colorectal cancer (CRC). Methods: Datasets from Gene Expression Omnibus (GEO) as well as The Cancer Genome Atlas (TCGA) were obtained to estimate the expression levels of ISM2 in CRC. ROC curve calculated the diagnostic value of ISM2. TIMER 2.0 was used to study the association between the ISM2 expression with immune cells and the main immune checkpoint. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses and Gene Ontology (GO) was further employed to study the related pathways. Finally, a couple of in-vitro and in-vivo assays, such as xenograft growth assay, were performed to verify the role of ISM2 in immune therapy and the progression of CRC. Results: ISM2 is highly expressed in CRC patients. Furthermore, ISM2 is a dependent risk factor. High expression of ISM2 is significantly associated with poor prognosis. More importantly, a couple of in-vitro and in-vivo assays verified the vital significance of ISM2 in the progression of CRC. Furthermore, the silence of ISM2 induced more CD8+ T cell infiltration in tumors and potentiated PD1ab therapy. Conclusion: ISM2 showed close association with clinicopathological features and tumor microenvironment in CRC.
