Water-Soluble Fraction from Nacre Suppresses Aging of Liver, Kidneys, and Brain of SAMP8 Mice

Nacre, also known as mother-of-pearl, has traditionally been used for health benefits. The aim of this study was to determine whether its administration could suppress amyloid-β accumulation, phosphorylated tau expression, and mitochondrial dysfunction associated with brain aging, as well as aging-related changes in the liver and kidney, using senescence-accelerated SAMP8 mice. The administration of water-soluble fraction from nacre attenuated memory decline and reduced amyloid deposition in the brain. It also significantly suppressed phosphorylated tau expression. Furthermore, the water-soluble fraction preserved mitochondrial function in the brain, preventing the age-related decline in ATP content and membrane potential in isolated mitochondria. Histological analysis of liver and kidney tissues revealed that the water-soluble fraction treatment suppressed fibrosis typically observed in SAMP8 mice. The expression of aging markers p21 and γ-H2AX, which were elevated in SAMP8 mice, was reduced by the water-soluble fraction treatment. Similarly, the age-associated increase in pro-inflammatory cytokines IL-1 and IL-6 was suppressed. Oxidative damage in liver and kidney tissues was also reduced in mice treated with the water-soluble fraction, and antioxidant activity in these tissues tended to be higher. These findings suggested that the water-soluble fraction from nacre administration mitigates not only brain aging, but also aging-related deterioration in the liver and kidneys of SAMP8 mice.
