A Comparative Physicochemical Study of Cortical and Cancellous Bovine Demineralized Bone Matrix

Demineralized bone matrix (DBM) provides a degradable matrix that releases endogenous bone healing compounds to the bone defect sites, inducing new bone formation and accelerating healing. The samples were collected from adult Bali cattle (Bos sondaicus), from which the diaphysis and metaphysis of the femur were taken as samples in the cortical and cancellous bone sections. The chemical properties were measured using Fourier Transform Infrared Spectroscopy (FTIR). The resistance capacity of each group was compared by giving axial force and measured using a Universal Testing Machine. The Acidity was measured daily using a pHmeter and degradation was measured one week after the sample was soaked in simulated Body Fluid (SBF). The FTIR analysis revealed significant differences in the composition of cortical and cancellous DBM, with cancellous DBM containing higher collagen and organic content, enhancing cell adhesion and osteoconductivity, while cortical DBM exhibited greater mineralization, contributing to mechanical strength and stability. The compression test in cortical DBM showed significantly higher results than in cancellous DBM. The acidity did not differ substantially during the 7-day evaluation. Meanwhile, the cancellous DBM is more dissolved (>50%) than the cortical DBM. The similarity of DBM was compared with carbonate substitution into the apatite lattice and collagen. The proportion between the mineral and organic bone content expressed its mineralization degree. The compression resistance, acidity, and degradability of demineralized bone matrix related to host and material interaction (local environment) promoted bone healing. The durability and biochemical properties mimicking the actual bone became a significant component of DBM usage in Bone Defect.
