Correlation of tumour necrosis factor-α -308 G/A polymorphism with schizophrenia in people with Malayan-mongoloid race in Western Indonesia

Schizophrenia is one of the most debilitating mental disorders that afflict mankind. A putative susceptibility locus for schizophrenia, the -308 G/A (rs361525) polymorphism, is situated in the promoter region of the TNF-α gene and has attracted substantial interest. Moreover, multiple studies have linked the A-allele to elevated TNF-α expression. It was essential to ascertain whether the promoter region influenced the inhabitants of Western Indonesia due to its paradoxical results. The subjects were adults of the Malayan-Mongoloid race, aged 15 to 40, diagnosed with schizophrenia according to the International Classification of Diseases, 10th edition criteria. They were required to be helpful and amenable to participating in an interview. The presence of mental problems, obesity, and general medical conditions affecting brain anatomy were deemed excluding factors. The inclusion criteria for the controls comprised individuals of the Malayan-Mongoloid race, aged 15 to 40, with no familial history of mental disorders, as well as a shown willingness to cooperate and participate in interviews. Obesity, general medical conditions affecting brain morphology, and a psychiatric disorder history were the exclusion criteria for the control group. Chi-square test and Hardy-Weinberg Equilibrium were used to analyse the result, and we found that there were no differences between genotypes and alleles in schizophrenia and healthy controls. In North Sumatera, we identified no correlation. between the -308 G/A TNF-α polymorphism and schizophrenia. A single polymorphism cannot be deemed a substantial risk factor for the onset of a specific disease.
