Effects of Lactiplantibacillus Plantarum IS-10506 on the Expression of Serotonin, Serotonin Transporter, and TNF-α in Sprague–Dawley Rat Ileum

Diarrhea is one of the important health concerns in children especially those in developing countries. Serotonin (5-HT) and 5-HT transporter (SERT) play critical roles in maintaining homeostasis of the gastrointestinal (GI) system, while tumor necrosis factor-alpha (TNF-α) contributes to immune regulation and inflammation. Probiotics, such as Lactiplantibacillus plantarum IS 10506, have potential therapeutic agents as they can modulate the gut microbiota, reduce inflammation, and restore gut homeostasis. This study investigated the effects of L. plantarum IS 10506 on the expression of 5-HT, SERT, and TNF-α in the ileum during Escherichia coli-induced diarrhea. This cross-sectional analytical observational study used ileum paraffin blocks from Rattus norvegicus (Sprague–Dawley strain). Ileal samples were divided into three groups: KN (placebo), KL (lipopolysaccharide [LPS] E. coli), and KP (LPS E. coli followed by L. plantarum IS 10506). LPS and L. plantarum were administered at 250 µg/kg body weight and at 2.86 × 1010 CFU, respectively. Immunohistochemical staining was conducted to evaluate 5-HT, SERT, and TNF-α expression and analyzed using normality tests and analysis of variance (p < 0.05). Ethical approval was obtained from the Faculty of Veterinary Medicine, Airlangga University. L. plantarum IS 10506 significantly modulated the expression of 5-HT (p = 0.029), SERT (p = 0.045), and TNF-α (p = 0.024) in the ileum. L. plantarum IS 10506 increases 5-HT and TNF-α expression following mucosal damage caused by E. coli LPS.
