Bioactivity of Pineapple Crown Fraction as an Antidiabetic Agent Targeting α-Glucosidase Inhibition and Increasing Glucose Intake in L6 Myoutubes of Rat

The ethanol extract showed antidiabetic activity expressed by the results of the inhibition analysis of α-glucosidase, α-amylase and glucose uptake in L6 myoutube rats. Ethanol extract was obtained from pineapple crown and fractionated to obtain 3 types of fractions, namely ethyl acetate fraction, n-hexane fraction, and water fraction. Each fraction was identified using H-NMR to determine the class of compounds present in it. H-NMR analysis of ethyl acetate, n-hexane, and water fractions showed the presence of phenolic compounds. Ethyl acetate fraction with a concentration of 250 µg/ml can inhibit α-glucosidase by 62.03% and α-amylase by 71.68%. Ethyl acetate fraction was able to increase glucose intake of L6 myoutube with a percentage increase of 89.82%. This figure is quite high when compared to insulin as a positive control. This study is the first report of pineapple crown fractionation, inhibition of α-glucosidase and α-amylase, and glucose uptake of L6 myoutube rats. Based on the results of this study, it was found that the ethyl acetate fraction of pineapple crown has antidiabetic activity.
