Solubility Enhancement of Metaxalone by Solid Dispersion Technique Using Polymeric Carriers

Limited aqueous solubility of New Chemical Entities (NCEs) has been one of the major challenges in pharmaceutics. Around 70% of the newly synthesized drugs fall into BCS (Biopharmaceutics Classification System) Class II (low solubility and high permeability). Therefore, solubility enhancement of poorly soluble drugs has been crucial to achieving greater aqueous solubility, and consequently bioavailability. Metaxalone is a skeletal muscle relaxant used in acute muscle pain in sprains and fractures. Metaxalone has low solubility and high permeability, categorizing it in BCS Class II. This project work aims to present the enhancement in aqueous solubility of Metaxalone, a poorly soluble drug, using the solid dispersion technique, particularly Solvent Evaporation method with the aid of polymers like PEG- 6000 and PVP K-30. MET/PEG- 6000 1:1 SDs showed approximately ~8-fold increase in aqueous solubility compared to pure drug.
