The Effect of Iron Nanoparticles Synthesized in the Rattle Plant on the Expression of Bax and BCL2 Genes

Background & Objective: Nanoparticles can improve the specificity of the treatment target, reduce the drug dose, maintain the effectiveness of the treatment and thus reduce the side effects of treatment. The present study was conducted with aim to investigate the effect of iron nanoparticles synthesized in the rattle plant on the expression of Bax and BCL2 genes and to evaluate the activity of caspases 8 and 3 in U87 ovarian cancer cells.Materials & Methods: In this study, the green synthesis of iron nanoparticles was done using rattle extract. The effect of nanoparticles in different concentrations and times was done using real time method.Results: The expression of BCL2 gene in the concentration of 200-100-50 mg decreased significantly and there was a significant difference between the gene expression in the concentration of 100 mg and BCL2 in the concentration of 200 mg/ml; while there was a significant difference between BAx gene expression at a concentration of 50 mg/ml and BAx at a concentration of 200 mg/ml. Iron particles with a concentration of 100-200 mg of nanoparticles were present in 48 hours after treatment. Also, the MT results of rattle extract showed that a significant difference between the concentration of 50 mg of the extract of the rattle plant after 72 hours of treatment with the concentration of 100 and 200 mg of the extract after 48 hours of treatment and also a significant difference between the concentrations of 200-400 and 600 mg of extract after 72 hours of treatment.Conclusion: In vitro conditions showed changes in the expression of pro-apoptotic BAX and anti-apoptotic Bcl2, which indicates a pro-apoptotic effect of all three components, especially iron nanoparticles containing rattle. Further clinical trials are required for the use of these nanoparticles in treatment of breast cancer.
