Flow Injection Spectrophotometric Method for Determination of Hesperidin in Pure and Supplement Forms

A cost-effective flow injection analysis (FIA) - spectrophotometric method has been devised for the simultaneous measurement of hesperidin (HES) in bulk powder and supplements. The method involves a diazotization coupling reaction of HES with diazotized 4-aminoacetophenone in an alkaline environment, leading to the formation of an orange azo-dye product with maximum absorption at 470 nm. Various chemical and physical parameters were optimized to enhance the sensitivity of the FIA procedure. The developed method exhibited a linear range of 5-100 µg/mL, with detection and quantification limits of 1.09 and 3.63 µg/mL, respectively, under optimal experimental conditions. Validation of the method demonstrated its reliability, with a low percentage relative standard deviation of less than 1.5% (n=5). Statistical comparisons using F and t-tests revealed negligible differences in accuracy and precision between the proposed FIA method and the traditional spectrophotometric approach.
