Highly Selective and Sensitive Voltammetric Aptasensor for Progesterone Determination in Different Samples

The blood level of progesterone (P4) is of vital importance in the reproduction health of mammalians. In addition, P4 level should be sufficiently low in food as it is suspicious to increase cancer risk. Therefore, various methods have been reported for selective and sensitive determination of P4. A new label-free aptasensor is designed for highly selective and sensitive analysis of P4 by using accessible and biocompatible materials. Direct oxidation of P4 was used as the signal probe. A glassy carbon electrode was modified with MWCNTs and AuNPs, then l-arginine was electropolymerized on AuNPs/MWCNTs/GCE by cyclic voltammetry. The P4 aptamer was deposited by using glutaraldehyde linker. The aptasensor showed a wide linear concentration range for P4 (0.12-345 nM or 0.028-99.7 ng/mL) and a limit of detection (LOD) of 0.12 nM or 0.028 ng/mL (based on S/N=3) by differential pulse voltammetry. The relative standard deviation (RSD%) for 6 replicate P4 measurements was 2.44%. For five similarly prepared electrodes, RSD% of P4 determination was 4.32%. The proposed aptasensor was applied in the determination of low P4 concentrations in human blood serum, cow milk and pharmaceuticals (injections), successfully.
