Syzigium aromaticum promotes Folliculogenesis and Alleviates Diminished Ovarian Reserve in a Rat Model

Background and objectives: Diminished ovarian reserve (DOR), as one of the reasons for female infertility, has been increasing worldwide with no specific medication. Syzygium aromaticum (clove) is mentioned for enhancing sexual performance and menstrual health in Persian medicine. The aim of the present study was to investigate the effect of clove extract on DOR induced by cyclophosphamide in rats. Methods: The clove extract was prepared using methanol 80%. Thirty female rats were randomly divided into five groups, including sham, DOR and three treatment groups (12.5, 25, and 50 mg/kg clove extract). DOR was induced in rats by intra-peritoneal injection of 45 mg/kg cyclophosphamide. Fourteen days after a single injection of cyclophosphamide, the rats were treated orally with clove extracts for 28 days. Subsequently, blood samples were collected for evaluation of anti-Müllerian hormone (AMH), estradiol (E2), and follicle-stimulating hormone (FSH); the ovaries were studied for histopathology. Results: The levels of AMH and E2 significantly decreased in the DOR group compared to the sham group, while FSH showed a significant increase. Clove treatment (12.5 mg/kg) increased AMH and decreased FSH compared to the DOR group, but E2 level remained unchanged. The administration of clove extract at the dose of 12.5 mg/kg increased the number of primordial, primary and antral follicles compared to the DOR group. However, the difference in cystic follicles, graffian and corpus luteum between the groups was not significant. Conclusion: Clove may be considered as a candidate for the treatment of DOR by regulating hormone responses and ovarian function.
