The Effect of Resveratrol on Cervical Cancer Cell Growth by Inhibiting TGF-β Gene Expression

Background & Objective: Resveratrol is a natural polyphenolic compound with anticancer and anti-inflammatory properties. Transforming Growth Factor Beta (TGF-β) plays a critical role in tumor growth and progression. This study was conducted with aim to investigate the modulatory effect of resveratrol on TGF-β gene expression in human cervical cancer cells (HeLa).Materials & Methods: HeLa cells were treated with standardized concentrations of resveratrol (50 μM and 100 μM). The IC50 value was determined, and total RNA was extracted for evaluation of TGF-β gene expression by real-time PCR. Apoptotic and cytotoxic effects were observed at 24, 48, 72, and 100 hours of treatment. Statistical analysis was performed using GraphPad Prism and one-way ANOVA, and Pearson's correlation tests. P<0.05 was considered statistically significant.Results: Resveratrol inhibited HeLa cell proliferation with an IC50 of 200 μM. Treatment significantly downregulated TGF-β gene expression (P<0.05). The strongest cytotoxic and apoptotic effects were observed at 48 and 100 hours, while a weaker response was noted at 96 hours. TGF-β expression peaked on day four but declined slightly with higher resveratrol concentrations.Conclusion: Resveratrol suppresses cervical cancer cell growth and downregulates TGF-β2 and TGF-βR2 mRNA expression in a time- and dose-dependent manner, supporting its potential as an adjunct therapeutic agent in cervical cancer
