Evaluation of Chemical and Biological Characteristics of Subcritical Water Extract from Helicteres isora L. Fruit and Its Incorporation into Chia Seed Mucilage Film

The purpose of this work was to assess the biological characteristic of subcritical water extract of Helicteres isora L fruit. Subcritical water extraction was carried out in particular circumstances (water temperature, flow velocity, pressure, process time, and sample mass were 175ºC, 4 mL/min, 120 min, and 4 g, respectively). Assessment of the chemical composition of H. isora fruit extract affirmed the existence of 2-propenoic acid, 2-ethylhexyl ester, heptadecen-(8)-carbonic acid-(1), isomenthol, 1-octen-3-ol, berberine, 6,10,14-trimethylpentadecane-2, docosanoic acid, 1,2,3-propanetriyl ester, phytol, formic acid,1-methylethyl ester, 2-methyl-1-butanol, hexadecanoic acid, and octadecnoic acid. At concentrations higher than 500, 200, and 100 µg/mL, H. isora fruit extract inhibited the growth Escherichia coli, Salmonella enterica, and Pseudomonas aeruginosa, respectively. The AGS cell line was found to be highly susceptible to H. isora fruit extract, with cell viability (%) of 41.1% at 1000µg/mL. The IC50 magnitude for MKN and AGS (tumor cell lines) were 58.9 ± 4.8 and 12.31 ± 1.3 µg/mL, respectively, while the IC50 for the normal cell line (HEK239) was 335 ± 28 µg/mL. The selectivity indices for AGS and MKN were equal to 27.21 and 5.68, respectively. Fabrication of active film based on chia seed mucilage and H. isora fruit extract showed inclusion of the extract influenced various properties of the resulted film. Thickness of films enhanced as greater magnitude of the H. isora fruit extract was the added (p<0.05).  The data depicted that the magnitude of TS and elongation in film samples enhanced as the H. isora fruit extract was incorporated into film formulation (p<0.05). The analysis of the antioxidant property of fabricated films affirmed that the addition of H. isora fruit extract had remarkably augment the antioxidant property of the film (p<0.05). The films possess H. isora fruit extract depicted bactericidal characteristic against Escherichia coli and Staphylococcus aureus.
