Synthesis of Small Molecule Biphenyl Carboxylic Acids as Anticancer Agents

A small molecule library of biphenyl carboxylic acids was synthesized and characterized by interpretation of 1H-NMR, 13C-NMR, Mass, and IR spectral data. The synthetic approach involved well-known Suzuki-Miyaura coupling catalysed by Pd (0), utilizing various substituted boronic acids. The new molecules were screened for their in vitro anticancer activity against MCF-7 and MDA-MB-231 breast cancer cells, by taking Tamoxifen as standard reference. Benzyloxy compound 3j displayed potent activity against both the cell lines with IC50 value of 9.92 ± 0.97 µM against MCF-7 and 9.54 ± 0.85 µM against MDA-MB-231 cell when compared to Tamoxifen. The other compound 3a with substituents were absent presented good activity with IC50 value of 10.14 ± 2.05 µM against MCF-7 and 10.78 ± 2.58 µM against MDA-MB-231. The potent molecule 3j tested in silico against crystal structure of estrogen receptor alpha revealed promising binding interactions with notable binding energy.
