The Intersection of Oncolytic Viral Therapy and Tumor Immunology: Boosting Prostate Cancer Immunotherapy

Because many tumors exhibit minimal immune infiltration, restricted antigen visibility, and a suppressive tumor microenvironment (TME), prostate cancer (PCa) is still challenging to sensitize to immunotherapy. By associating specific tumor-cell infection with immunogenic cell death, antigen release, and innate immune activation, oncolytic viruses (OVs) may be able to overcome this restriction. When utilizing with immune checkpoint inhibitors (ICIs), these medicines may help transform immune-excluded illness into a more checkpoint-responsive state by promoting OV-mediated inflammation. In order to convert OV-ICI combinations into clinically significant PCa immunotherapy, this editorial contends that logical viral engineering, biomarker-guided selection, and cautious control of antiviral barriers are crucial.
